Abstract

The preparation and characterization of gold nanoparticles in white turmeric extract (Curcuma zedoaria (Berg.) Roscoe) with various extract concentration aims to determine the optimal extract concentration and its effect on particle size, PDI, % EE, and potential zeta of the formed nanoparticles. Nanoparticles are made by green synthesis method by utilizing phenolic compounds and flavonoids which have antioxidant activity from the extract as a reducing agent and stabilizer of nanoparticle synthesis. The variations in extract concentration are 10 mg, 30 mg, and 90 mg. The optimal concentration needed to obtain the optimum formula is 30 mg. The greater the concentration causes the faster the nanoparticle increases, the nanoparticle decreases, and the more potential to increase agglomeration. The optimum formula of nanoparticles obtained has % EE value of 91.723% ± 0.092 with a time of 1.11 minutes to change the solution to purple which indicates the formation of gold nanoparticles. The characterization result of the optimum nanoparticle formula has a diameter of 61.34 nm, a PDI of 0.396 and a zeta potential of -27.2 mV. The physical stability of the nanoparticles optimum formula by the heating cooling test method produces organoleptic form of a purple solution with a cluster formed and pH value of 6.193 ± 0.006. The antioxidant activity test the by DPPH method (2,2-diphenyl-1 picrylhydrazyl) obtained IC50 values of white turmeric extract, and the optimum formula of gold nanoparticles in white turmeric extract are 16,2682 ppm and 25,4608 ppm, respectively. These results indicate white turmeric extract has stronger antioxidant activity compared to the optimum formula of gold nanoparticles in white turmeric extract.
