Abstract

Daun ketepeng cina (Cassia alata Linn.) dapat digunakan sebagai obat secara tradisional. Daun ketepeng cina (Cassia alata Linn.) mengandung senyawa metabolit sekunder seperti flavonoid, alkaloid, fenolik, saponin, steroid, terpenoid, dan tanin dimana senyawa metabolit sekunder tersebut diduga dapat berkhasiat sebagai hepatoprotektor. Tujuan dari penelitian ini untuk mengetahui kemampuan ekstrak etanol daun ketepeng cina (Cassia alata Linn.) sebagai hepatoprotektor dan untuk mengetahui histopatologi hepar pada tikus putih jantan galur Wistar yang terinduksi karbon tetraklorida (CCl4). Parameter yang digunakan dalam penelitian ini yakni pengukuran kadar SGPT dan kadar SGOT serta gambaran histopatologi hepar. Tikus dibagi menjadi enam kelompok yaitu kelompok kontrol normal (tanpa perlakuan), kelompok kontrol positif (silimarin 200 mg/kgBB), kelompok kontrol negatif (CCl4 0,5 mL/kgBB), kelompok uji ekstrak etanol daun ketepeng cina (Cassia alata Linn.) dosis 100, 200 dan 400 mg/kgBB. Kadar SGPT dan kadar SGOT diukur dengan metode enzimatis. Variasi dosis ekstrak etanol daun ketepeng cina (Cassia alata Linn.) berpengaruh dalam penurunan kadar SGPT dan SGOT pada tikus putih jantan galur wistar yang diinduksi karbon tetraklorida (CCl4). Semakin besar dosis maka semakin besar efek penurunan kadar SGPT dan kadar SGOT (p>0,05). Hasil gambaran dari histopatologi hepar pada kelompok normal, negatif, positif, uji I, uji II dan uji III berupa kondisi nekrosis secara berturut-turut yaitu 0%, 80%, 1%, 20%, 15% dan 2%. Ekstrak etanol daun ketepeng cina (Cassia alata Linn.) dengan dosis 400 mg/kgBB (uji III) memberikan gambaran aktivitas hepatoprotektor yang paling baik dibandingkan dosis 100 mg/kgBB (uji I) dan dosis 200 mg/kgBB (uji II). Berdasarkan hubungan persen hepatoprotektor terhadap dosis ekstrak, maka nilai dosis efektif 50 (ED50) ekstrak etanol daun ketepeng cina (Cassia alata Linn.) sebesar 53,73mg/kgBB.
