Abstract

Methylprednisolone, used as therapy to suppress inflammatory reactions and skin allergies. This study aims to formulate ethosome methylprednisolone which has good penetration on topical. Ethosome were prepared by cold method and characterized for pH, viscosity, and percent efficiency entrapment. The optimization method used for ethosome methylprednisolone was 23 factorial design. The optimum formula was obtained at concentration soya lechitin 1%, propylene glycol 5%, and ethanol 96% 40%. Percent efficiency entrapment was conducted to find out how much drug entrapment in the vesicle and the result shows that vesicle contains 84.849±0.059% with %CV value of 0.059% which means that result obtained uniform. Diffusion test in vitro was performed using Franz diffusion cell conducted to determine the power penetration ethosome, cream marketed, and pure methylprednisolone. From the in vitro drug diffusion analysis, it was found that 24.975±0.752% of ethosome, marketed cream 12.760±1.545%, and pure methylprednisolone only penetrated 7.171±4.216%. It shows significant difference between penetration ability of ethosome, marketed cream, and pure methylprednisolone with p-value <0.05. Ethosome vesicle are to be included in the submicro size based on PSA measurements of 533,6 nm. The value of PI (polydispersity index) obtained <0.5 ie 0.193 which means that result has a homogen distribution. Potential zeta test is performed to determine the charge from the surface of the particle, obtained potential zeta value close to 25mV of -19.4 mV which means that result no aggregation on the suspension. The stability of the ethosome is relatively better than the marketed cream and pure methylprednisolone. Based on the percentage efficiency of entrapment and diffusion test, ethosome feasible to develop, but ethosome vesicles has a poor stability because it is easy for the occurance of precipitation.
