Abstract

Rubber is one of the agricultural commodities that play an important role in the economy of society and the country both internationally and specifically in Indonesia. Rubber plants continue to be developed in order to increase optimal production results. However, there are many obstacles in development, one of which is deciduous leaves caused by Pestalotiopsis sp. This study aims to explore endophytic bacteria to determine the effectiveness of endophytic bacteria in suppressing fall leaf pathogens Pestalotiopsis sp. The research was carried out at the Sembawa Research Institute in the Plant Protection laboratory and in the Rubber District of Banyuasin, South Sumatra Province, which was conducted in September 2019 to January 2020. The study was carried out by taking samples of leaves, stems and roots of rubber plants from the garden of budwood nursery and mature plant for suspension. The suspension is made in a dilution series of up to 10-4. Dilutions of 10-2 up to 10-4 each taken 1ml of suspension and cultured on Potato dextrose Agar (PDA) media with 3 replications. In vitro antagonistic test of endophytic bacteria by Pestalotiopsis sp was carried out by the dual culture method. Observations were made by measuring the fingers of Pestalotiopsis sp away from endophytic bacteria and fingers approaching endophytic bacteria. Endophytic bacterial in planta antagonist test with Pestalotiopsis sp was carried out with a 24 hour incubation preventive method by inoculating endophytic bacteria on rubber leaves and pathogen Pestalotiopsis sp. The results of exploration of endophytic bacteria found 25 isolates, 3 of which have antagonistic ability against Pestalotiopsis sp. The identification results showed that the 3 bacteria were Bacillus sp1, Pseudomonas sp and Bacillus sp2. The antagonistic mechanism carried out by Bacillus sp1 and Bacillus sp2 is antibiosis, lysis and space competition. Whereas Psudomonas sp is antibiosis and space competition.
