Abstract

This study aims to determine the effect of heating medium on Enterococcus faecalis heat resistance. This research method uses randomized block design (RBD) with one treatment and 3 replications. The treatment used is the difference in heating medium (rusip, bekasam and physiological salt). The first step of the research was media characterization, the results showed that different heating medium had significantly different pH value (4.41-6.99), water content (72.56-98.09%), ash content (0.69-12.32%), protein content (0-14.7%), fat content (0-0.84%) and carbohydrate content (0-4.93%). The characterized media were used for the second stage of research, namely heating Enterococcus faecalis at a temperature of 70oC for 0, 2, 4, 8 and 12 minutes. The number of surviving bacterial colonies was calculated using the TPC (Total Plate Count) method. The results of bacterial survival analysis showed that heating medium had no significant effect on bacterial heat resistance. The average value of bacterial log reduction per minute in bekasam, rusip and physiological salt media were 0.28 log cfu/min, 0.32 log cfu/min and 0.38 log cfu/min. The chemical compounds in bekasam can protect Enterococcus faecalis cells from damage during heating as indicated by lowest log reduction per minute.
