Abstract
Wistar Strain Male Rats with Fixed-Dose-Procedure Method. Mona Riska Andhini 08061381520037 ABSTRACT Robusta coffee leaf (Coffea canehora) is one of the plants used by the community as antidiabetic. Aside from being an antidiabetic, other properties are antimicrobial and antioxidant. The results of chemical composition screening containing coffee leaf substances contain alkaloids, flavonoids, phenolics, tannins, terpenoids and steroids. Chemical compounds contained in powerful robusta coffee leaves need safety drugs that are supported by toxicity testing. In this research, the toxicity test of the ethanol extract of Robusta (Coffea canephora) ethanol extract on Wistar strain male rats was carried out using the fixed dose procedure method. The ethanol extract of Robusta coffee leaves was obtained by meseration using 96% ethanol. The initial dose chosen for the main test is determined from the results of the preliminary test. Based on preliminary test results, a dose of 2000 mg / kgBW was determined as the dose in the main test. In the main test, the test animals were divided into 2 groups, namely the normal group and the 2000 mg / kgBB dose group, each consisting of 5 test animals. The results of observations in the main test showed that in the normal group and the dose group of 2000 mg / kgBW there were no deaths or toxic effects in the test animals. The range of toxic dosages of Robusta coffee leaf extract is> 2000 mg / kgBB. Ethanol extract of Robusta coffee leaves did not fight macroscopic renal, liver and cardiac test animals (р> 0.05). The average level of biochemical parameters of the normal group was SGOT 102.83 ± 26.00 U / L, SGPT 88.15 ± 17.27 U / L, creatinine 0.67 ± 0.09 mg / dL and ureum 18.01 ± 2 , 24 mg / dL. Whereas the dose group of 2000 mg / kgBW were SGOT 114.50 ± 31.48 U / L, SGPT 106.65 ± 36.16 U / L, creatinine 1.20 ± 1.0 mg / dL and ureum 36.40 ± 3 .07 mg / dL. Mona Riska Andhini 08061381520037 ABSTRAK Daun kopi robusta (Coffea canehora) merupakan salah satu tanaman yang digunakan oleh masyarakat sebagai antidiabetes. Selain sebagai antidiabetes, khasiat lainnya adalah sebagai antimikroba dan antioksidan. Hasil skrining kandungan kimia menunjukkan bahwa daun kopi robusta mengandung senyawa berupa alkaloid, flavonoid, fenolik, tannin, terpenoid dan steroid. Senyawa kimia yang terkandung di dalam daun kopi robusta yang berkhasiat sebagai obat perlu diketahui keamanan penggunaannya yang didukung oleh pengujian toksisitas. Pada penelitian ini telah dilakukan pengujian toksisitas akut ekstrak etanol daun kopi robusta (Coffea canephora) terhadap tikus jantan galur wistar dengan metode fixed dose procedure. Ekstrak etanol daun kopi robusta diperoleh dengan cara meserasi menggunakan etanol 96%. Dosis awal yang dipilih untuk uji utama ditentukan dari hasil uji pendahuluan. Berdasarkan hasil uji pendahuluan, dosis 2000 mg/kgBB ditetapkan sebagai dosis pada uji utama. Pada uji utama hewan uji dibagi menjadi 2 kelompok yaitu kelompok normal dan kelompok dosis 2000 mg/kgBB yang masing-masing terdiri dari 5 ekor hewan uji. Hasil pengamatan pada uji utama menunjukkan bahwa pada kelompok normal dan kelompok dosis 2000 mg/kgBB tidak terjadi kematian maupun gejala toksis pada hewan uji. Rentang dosis toksis ekstrak daun kopi robusta adalah dosis >2000 mg/kgBB. Ekstrak etanol daun kopi robusta tidak berpengaruh terhadap makroskopis ginjal, hati dan jantung hewan uji (р>0,05). Rata-rata kadar parameter biokimia kelompok normal SGOT 102,83±26,00 U/L, SGPT 88,15±17,27 U/L, kreatinin 0,67±0,09 mg/dL dan ureum 18,01±2,24 mg/dL. Sedangkan kelompok dosis 2000 mg/kgBB adalah SGOT 114,50±31,48 U/L, SGPT 106,65±36,16 U/L, kreatinin 1,20±1,0 mg/dL dan ureum 36,40±3,07 mg/dL.
