Abstract

Hyperuricemia is a condition where the level of uric acid in the blood is greater than normal, a disease caused by hyperuricemia is known as gout. One of the plants that can be used as antihyperuricemia is jengkol (Archidendron jiringa) leaves. Jengkol leaves contain flavonoid compounds such as kaempferol and quercetin which have decreased uric acid activity. Inhibition of the action of the xanthine oxidase enzyme by flavonoid compounds kaempferol and quercetin causes inhibition of the change in hypoxanthine to xanthine, which in turn converts xanthine to uric acid. So that it can reduce the production of gout. This study aims to determine the antihyperuricemia activity of ethanol extract jengkol leaves in white male rats induced by potassium oxonate. Used 25 white male rats (Rattus norvergicus) Wistar strain aged 2-3 months with a body weight of 150-250 grams. Test animals were divided into 5 groups with each group consisting of 5 rats namely positive control (allopurinol 3,941mg / 200gBW), negative control (Na CMC 0.5%), group I (250 mg / kgBW), group II (500 mg / kgBW) and group III (750 mg / kgBW). All test animals were induced with 4,926 mg / 200gBW of potassium oxonate intraperitoneally and 3 mL / 200 gBB of chicken liver juice orally for seven days to achieve hyperuricemia. Then the rats were given test preparations for seven days. The results showed the percentage of decreased uric acid levels in groups with a dose of 250 mg / kgBW, 500 mg / kgBW, and 750 mg / kgBB by 31.34%, 47.74%, and 66.80%. Based on the results obtained, it can be seen that the ethanol extract jengkol leaves has activity as an antihyperuricemia and there is a significant difference compared to the negative control group (p <0.05). The effective dose 50 (ED50) obtained from the equation between the dose and percent reduction in uric acid levels (% PKAU) of ethanol extract jengkol leaves was 526.29 mg / kgBW.
