Abstract
The in vivo antidiarrheal activity test of the combination of n-hexane and ethyl acetate fraction of karamunting leaves (Rhodomyrtus tomentosa (Ait.) Hassk.) against Salmonella typhi bacteria was carried out in order to determine the standardization of n-hexane fraction of karamunting leaves and to determine the optimum dose of combination n-hexane and ethyl acetate fractions of karamunting leaves as antidiarrheal. The animal test used male white rats Wistar strains which were divided into 5 groups consisting of a positive control group using a chloramphenicol dose of 25 mg/kg BW, negative control group using 1% Na CMC suspension and a combination treatment group of n-hexane and ethyl acetate fractions of karamunting leaves with variations in the doses of 25, 50, 100 mg/kg BW. The results of the standardization of the n-hexane fraction showed that the moisture content 6.67%; drying loss 7.16%; total ash content 0.95%; acid insoluble ash content 0.61%; water soluble extract content of 3.34%; ethanol soluble extract content 31.67%; microbial contamination test <10 cfu/g; Pb metal contamination test 1.4606 mg/kg; and Cd metal contamination test 0.2189 mg/kg. The fastest returned of feces consistency at dosage combined fraction of 100 mg/kg BW on the 6th day. The combination of fractions at a dose of 25 mg/kg BW and 50 mg/kg BW was able to reduce the number of bacterial colonies to 2.72 x 106 cfu/g and 1.58 x 106 cfu/g, less than the negative control 5.64 x 106 cfu/g. The combination of fractions at a dose of 100 mg/kg BW can reduce the Salmonella typhi bacterial population to zero after 12 days of treatment. This shows that the combination of n-hexane and ethyl acetate fractions of karamunting leaves has better antidiarrheal activity at the highest dose (100 mg/kg BW) and there is no significant difference compared to chloramphenicol at a dose of 25 mg/kg BW (p>0,05).
