Abstract
The distribution of giant snakehead (Channa micropeltes) in Indonesia spread in Southeast Sumatra, West Kalimantan, Java and Bangka Belitung. In South Sumatra, giant snakehead is found commonly in the Musi and Kelekar River. The purpose of this study was to determine the sequence of the mitochondrial DNA cytochromic C oxidase subunit I (COI) gene, the phylogenetic tree and genetic distance and determine the water chemical and physics of the of giant snakehead habitat. The methods used in species barcoding and sequence variation were DNA isolation, electrophoresis, amplification using PCR (Polymerase Chain Reaction) and sequencing of COI mtDNA gene region from fish samples obtained from the Musi and Kelekar rivers. This research was conducted from March – July 2020. The length of COI gene of common giant snakehead nucleotides was aproximately 716 bp. The results of BLAST analysis showed that giant snakehead had the highest similarity with the DNA sequences of giant snakehead originating from Switzerland (MF496867), which was 99.84% and the lowest similarity 95.28 % with giant snakehead from Malaysia (JF900369). The water qualites in the Kelekar and Musi River were temperature 29.1-31.0ºC, pH 6-7.6, dissolved oxygen 5.3-7.64 mg/L, alkalinity 12-36 mg/L, brightness 23-40 cm, ammonia 0.29-0.8 mg/L, water velocity 0.15-0.40 m/sec and TDS 23-27 mg/L.
