Abstract

Marble goby (Oxyeleotris marmorata) is distributed in Indonesia, mainly found in Sumatra, Kalimantan and Papua. The purpose of this study was to obtain the sequence of mitochondrial DNA Cytochrome Oxidase Subunit I gene, construct the phylogenetic tree and genetic distance between populations and determine the physical and chemical characteristics of the waters. This research was carried out in March - August 2020. The methods used in barcoding species and determining genetic relationships were DNA isolation, amplification method using PCR (Polymerase Chain Reaction) and sequencing of the COI mtDNA gene from domestication of marble goby in Gandus Fish Seed Center (BBI), Palembang City and the wild species from Musi River in Beruge Village, Babat Toman District, Musi Banyuasin Regency. The sequence of COI mtDNA gene fragments were obtained from PCR results with an annealing temperature optimization of 50°C for 30 seconds in 35 cycles. After going through the editing process, the nucleotides length of COI gene of marble goby were 613 basepairs. Based on the BLASTn (Basic Local Alignment Search Tool nucleotide) analysis with comparisons of data in GenBank, domesticated marble goby (O. marmorata) (OMD2, OMD3) was in the same subcluster with O. marmorata from the Musi River (OMS2, OMS3). Domesticated marble goby (OMD2, OMD3) and the wild fish of marble goby from the Musi River (OMS2, OMS3) had a 99.35% similarity with O. marmorata from Cambodia (EF609424.1), and Indonesia (West Java) (KU692718.1 and KU692726.1) with a genetic distance of 0.002 (0.02%). The characteristics of O. marmorata in BBI Gandus and Musi River were temperature 29.1-30.9oC, water transparency 13-26 cm, dissolved oxygen 6.5-7.75 mg L-1 , pH 6, ammonia 0.01-1.53 mg L-1 , TDS 14-25 mg L-1 and total alkalinity 16-21 mg L-1 CaCO3.
