Abstract

A research regarding formulation and optimization of gel from erythromycin submicro particles using gelling agent HPMC 60-SH has been conducted. The concentration of HPMC 60-SH was used 2% for first formula, 4% for second formula, and 6% for third formula. Based of the observation result showed that HPMC 60-SH 2% has been produced a gel with highest quality than any other formula. The test result of viscosity, adhesibilty, spreadbility, washability, pH, and extrudability from formula using HPMC 60-SH 2% were by 1111 ± 13,75 cP; 69,23 ± 1 detik ; 5,27 ± 0,11 cm; 13,67 ± 0,005 mL; 5,35 ± 0,005; 2,42 ± 0,09 cm. Gel of HPMC 60-SH 2% showed good homogeneity and free of agglomerates. Its stability is good because the organoleptic, pH, and homogeneity remain stable during last cycle of heating cooling. The effectiveness of gel was observed through in-vivo testing with animal test infected to Staphylococcus aureus. Number of lesions on test animal skin smeared with gel from erythromycin submicro particles were lost on the 9th day, whereas in the other group of test animals smeared with optimum formula without active substances, HPMC plus 50 mg erythromycin, and without treatment still showed the presence of red lesions on the surface of test animal skin on the 9th day. Gel from erythromycin submicro particles with HPMC 60-SH 2% showed the greatest and fastest decrease of lesions when compared with other group of test animals.
