Abstract

Ludwigia peruviana is one of the 82 species of Ludwigia that grows in the tropical and sub-tropical. The extract of L. adscendens has been reported as antidiabetic activity. Meanwhile, the antidiabetic activity and the chemical compound isolated from L. peruviana has never been reported yet. Generally, the same genus have the similar compound. Therefore, the research was conducted to isolated the compound from the stem L. peruviana and antidiabetic assay using α-glukosidase enzyme from the methanol extract, n-hexane and ethyl acetate fraction as well as the isolated compound. The extraction from the stem of L. peruviana was done by maceration with methanol as the solvent, continuing with liquid-liquid partition using n-hexane and followed by ethyl acetate. The separation and purification was performed using vacum liquid chromatography, column sephadex (LH-20), and gravity chromatography. The Identification of the isolated compound was determined by infra red spectroscopy and gas chromatography-mass spectra. The Isolate compound was produces as white solid (19.8 mg). The spectrospocy data showed that the isolated compound was a mixture of the three steroid compounds, which the major compounds was β-sitosterol. The antidiabetic assay using α-glukosidase enzyme from the methanol extract, n-hexane fraction, ethyl acetate fraction , and the isolated compound showed the value of IC50 were 59.23, 73.96, 62.52, and 283.94 ppm respectively. Meanwhile the IC50 value of acarbose as positive control was 80,77 ppm. The IC50 data showed that the methanol extract and ethyl acetate fraction are stronger antiabetic ability than the n-hexane fraction and isolated compound
