Abstract

Ubi Jalar Ungu (Ipomoea batatas L. Poir) merupakan tanaman yang sering digunakan oleh masyarakat sebagai bahan pokok yang sering digunakan masyarakat. Daun ubi jalar ungu dalam ekstrak n-heksana diduga mengandung triterpenoid berupa betakaroten yang dapat berkhasiat sebagai hepatoprotektor. Penelitian ini betujuan untuk melihat aktivitas ekstrak n-heksana daun ubi jalar ungu sebagai hepatoprotektor dengan melihat kadar SGPT dan SGOT, gambaran makroskopis dan mikroskopis hati tikus putih jantan galur Wistar. Sebanyak 30 tikus dibagi menjadi enam kelompok dalam 16 hari pengujian yaitu kelompok I kontrol normal (NaCMC 1%), kelompok II kontrol negatif (Aloksan 125 mg/kgBB), kelompok III kontrol positif (silimarin 200 mg/kgBB), serta kelompok perlakuan ekstrak n-heksana daun ubi jalar ungu dosis 250, 500, dan 1000 mg/kgBB. Pengambilan darah tikus dilakukan setelah diinduksi aloksan dan setelah dilakukan perlakuan. Tikus selanjutnya dikorbankan dan dilakukan pengamatan secara makroskopis dan mikroskopis pada organ hati. Hasil penelitian menunjukkan bahwa ekstrak n-heksana daun ubi jalar ungu dapat berkhasiat sebagai hepatoprotektor dengan % efek hepatoprotektor (% EH) untuk kadar SGPT dan SGOT pada kelompok uji I sebesar 33,23% dan 28,57%, kelompok uji II sebesar 48,23% dan 46,616%, dan kelompok uji III sebesar 88,43% dan 79,55%. Berdasarkan hubungan persen hepatoprotektor terhadap dosis ekstrak, maka nilai dosis efektif 50 (ED50) ekstrak n-heksana daun ubi jalar ungu SGPT dan SGOT sebesar 449,03 dan 553,58 mg/kgBB. Gambaran makroskopis organ hati kelompok uji sesuai dengan kriteria hati normal. Gambaran mikroskopis hati kelompok uji memiliki skor kerusakan lebih kecil dibandingkan dengan kelompok kontrol negatif tanpa obat. Hasil pemberian ekstrak n-heksana daun ubi jalar ungu dengan dosis 250, 500, dan 1000 mg/kgBB (p>0,05) memiliki pengaruh dalam aktivitas hepatoprotektor. Dosis 500 mg/kgBB memberikan aktivitas hepatoprotektor yang lebih baik dibandingkan dengan dosis 250 dan 1000 mg/kgBB.
