Abstract

This study aims to determine the effectiveness of red spinach leaf extract (Amaranthus tricolor L) for coloring mango (Mangifera indica L) stem tissue preparations and to find the optimum concentration that can color the mango stem tissue preparations as a media for learning biology. This type of research is real experiment and analyzed by means of descriptive qualitative. Preparations were made using the method without paraffin, with various concentrations, namely (20%, 40%, 60%, 80%, 100%). The research was conducted in the Biology Education Laboratory of Palembang, Sriwijaya University. The results of this study indicate that the red spinach leaf extract can color the mango stem tissue preparations with very good criteria at a concentration of 100% because the color of the cell walls and cytoplasm of each cell is bright or bright. However, the results showed that to differentiate between one cell and another was not only affected by good staining but also strongly associated with the thin incision of the preparations.
