Abstract

A research regarding formulation and optimization of gel from ketoconazole submicro particles using gelling agent Carbopol®934 and HPMC 60-SH has been conducted. The concentration of Carbopol®934 was used 1% for first formula and 2% for second formula, while HPMC 60-SH was used 3% for third formula and 6% for fourth formula. Based of the observation result showed that Carbopol®934 2% has been produced a gel with highest quality than any other formula. The test result of viscosity, adhesibilty, spreadbility, washability, pH, and extrudability from formula using Carbopol®934 2% were by 1208±45.43 cP; 24.00±1.00 detik; 6.22±0.05 cm; 6.76±0.25 mL; 4.69±0.01; dan 2.63±0.12 cm. Gel of Carbopol®934 2% showed good homogeneity and free of agglomerates. Its stability is good because the organoleptic, pH, and homogeneity remain stable during last cycle of heating cooling. Gel testing evaluation showed there was no irritation on test animal. The effectiveness of gel was observed through in-vivo testing with animal test infected to Trichophyton sp. Number of lesions on test animal skin smeared with gel from ketoconazole submicro particles were lost on the 10th day, whereas in the other group of test animals smeared with submicro particles, commercial ketoconazole, placebo, and without treatment still showed the presence of red lesions on the surface of test animal skin on the 15th day. Gel from ketoconazole submicro particles with Carbopol®934 2% showed the greatest and fastest decrease of lesions when compared with other group of test animals.
