Abstract

Petai skin (Parkia speciosa Hassk.) is a plant that has immunomodulatory activity. Medicinal plants whose properties have been known must go through a toxicity test to determine their level of safety, one of which is a subchronic toxicity test. In this study, a subchronic toxicity test of the ethanol extract of petai skin (Parkia speciosa Hassk.) was carried out on male and female white rats Wistar strain. This study used 25 male rats and 25 female rats which were divided into 5 groups, consisting of a control group and 4 treatment groups with doses of 100, 200, 400, and 800 mg/kgBW. This study lasted for 28 days. The parameters observed included body weight, macroscopic observations, measurement of blood biochemical parameters (SGOT, SGPT, creatinine, urea, blood glucose and cholesterol) and blood hematology parameters (hemoglobin, erythrocyte, leukocytes and hematocrit) which were carried out on day 0 and day 29th. The results showed a change in the weight of the tested animals before and after treatment. In the control group male rats, 100 mg/kgBW, 200 mg/kgBW, 400 mg/kgBW, and 800 mg/kgBW increased respectively by 46.52%; 43.42%; 38.98%; 51.8%; and 50.07% and in the control group female rats, 100 mg/kgBW, 200 mg/kgBW, 400 mg/kgBW, and 800 mg/kgBW respectively increased by 37.07%; 31.47%; 39.77%; 38.59%; 34.8%. Test of blood biochemical parameters (SGOT, SGPT, creatinine, urea, blood glucose and cholesterol) and blood hematological parameters (hemoglobin, erythrocyte, leukocyte and hematocrit) for all treatment doses showed values that were still within normal limits. Because if there is a toxic effect, there will be an increase that exceeds the normal limit for each parameter. Based on these data, it can be concluded that the ethanol extract of petai skin (Parkia speciosa Hassk.) is not subchronic toxic up to a dose of 800 mg/kgBW.
