Abstract

Transfersome is able to facilitate the penetration of drugs into the stratum corneum of the skin. Vesicles can entrap pharmaceutical agents that are hydrophilic such as flavonoid. The transfersome formulation of ethanol extract of the Helmithostachys zeylanica (Linn) Hook rhizome uses a thin-film hydration method with a difference of surfactant concentration (tween 80); within the range 5 - 10% w/v. Soya lecithin is used as the main component of transfersome forming, with phosphate buffer pH 7.4 as buffering agent. The qualitative test of flavonoid content with thin layer chromatography using silica gel plate GF245 and the developer of ethyl acetate: n-hexane (40:65) has an Rf value of 0.37 and 0.72. Purfersome purification by centrifugation method at 14,000 rpm, 30 min, 4°C. The determination of transfersome optimum formula is seen from the characteristics of the best formula of the three formulas, which are three batches. Formula 2 was chosen as the optimum formula because it has a value of %EE up to 63.004 ± 0.920%. Transfersome characteristic of optimum formula is white sphere, particle diameter 871.5 nm, zeta potential 27.3 mV, value of PDI 0.489, viscosity 0.600 ± 0.050 dPa.S. The total flavonoid content of ethanol Helmithostachys zeylanica (Linn) Hook rhizome point was 261.667 mg/g and total flavonoids total transfersome optimum formula up to 164.861 ± 2.406 mg/g at λmaks 510 nm. The cycling test stability test at 4°C and 40°C indicates that the preparation is stable at low temperature (4°C) and unstable at high temperature (40°C). Different concentrations of tween 80 had an effect on initial pH, viscosity and percent encapsulation (% EE), but had no effect on final pH after stability test of cycling test.
