Abstract
Azithromycin is a macrolide antibiotic belongs to BCS class II with poor solubility properties causing extremely low dissolution in gastrointestinal tract and lower bioavailability. Development into SNEDDS dosage form has effectively known to improve solubilization of hydrophobic drug. Capryol-90 was used as oil phase in SNEDDS as it can dissolved azithromycin properly (1.096 ±0.028 mg/mL). Tween-80 and PEG-400 was used as non-ionic surfactant and cosurfactant due to have an high HLB value (>12) in order to increase azithromycin solubility in water. Determination of ternary phase diagrams conducted to obtain proporsion of combination component formed spontaneous nanoemulsion region. Nanoemulsion region formed on concentration range of capryol-90 10 – 60 %,tween-80 20 – 50 %, and PEG-400 10 – 70 %. Determination of 13 SNEDDS azithromycin formulas used simplex-lattice-design method in Design-Expert®10 program. Component proportion of SNEDDS azithromycin optimum formula from optimization process toward 7 characteristics of 13 formulas resulted 20.29% capryol-90, 48.85% tween-80, and 30.86% PEG-400. Optimum formula resulted characteristics percent of transmittance (83.92 ± 1.45)%, emulsification time (107.31 ± 5.03) second, viscocity (68.75 ± 4.68) cP, pH (7.89 ± 0.04),robustness to dilution in aquadest, SGF, and SIF (99.84 ± 0.12)%, (98.9 ± 0.42)%,dan (99.7 ± 0.30)%. Dissolution test showed that SNEDDS can improve azithromycin dissolution in SIF pH 7.4 compared to pure, and generic tablet azithromycin with increased SNEDDS’s DE60 value up to (90.87 ± 0.67)%, while pure azithromycin (5.38 ± 0.52)%, and generic tablet (6.14 ± 0.80)%. SNEDDS azithromycin resulted particle size 194.3 nm, has a good uniformity (PDI 0.095),but has unstable tendency due to surface charge value of – 4.9 mV. Based on results in this research, azithromycin can be prepared into SNEDDS dosage form.
