Abstract

Penelitian ini bertujuan untuk memperoleh isolat bakteri asam laktat (BAL) tahan panas asal bekasam. Penelitian ini dilaksanakan pada Mei hingga Agustus 2018. Metode penelitian ini menggunakan ekspremental laboratoris dan analis data dilakukan secara deskriptif. Tahap penelitian meliputi proses pengambilan sampel uji, pemanasan bekasam, uji Total Plate Count, isolasi bakteri asam laktat, regenerasi bakteri, uji konfirmasi bakteri asam laktat (uji pewarnaan gram, uji katalase dan uji motilitas), uji ketahanan terhadap suhu dan waktu pemanasaan pada suhu 70, 75, 80, 85 dan 90 oC selama 2, 4, 6, 8 dan 10 menit. Hasil penelitian ini menunjukkan bahwa jumlah koloni BAL pada sampel BT1, BT2 dan BT3 bekasam sebanyak 5,30 log Cfu/mL, 6,20 log Cfu/mL dan 6,37 log Cfu/mL. Sampel BP1, BP2 dan BP3 pasteurisasi sebanyak 4,10 log Cfu/mL, 4,09 log Cfu/mL dan 4,25 log Cfu/mL. Sampel BP1, BP2, BP3 menghasilkan 12 isolat BAL 5 diantaranya merupakan kandidat BAL tahan panas yaitu B2, B4, B5, B7 dan B10. Semua isolat kandidat BAL mampu bertahan pada pemanasan 90 oC.
