Abstract

Pak choi is a high valued leaves vegetables that require an optimal nutrient to enhance its productivity. The relatively low soil quality and the attack of pest couse a decrease in the productivity of pak choi as a crop in Indonesia. This research aims to explore and characterize the morphology, physiology and moleculer identity of PGPR bacteria contained in use maggot compost and use maggot isolate collection from Agricultural Microbiology Lab. Pusat Penelitian Biologi LIPI, and to see teh effects of the bacteria to inhibits the pathogenic activity of Fusarium oxysporum and increase the growth rate of pak choi. The research has done in Agricultural Microbiology Lab. LIPI from June 2020 to January 2021. Isolation from 4 use maggot compost sample has done with a different concentration; which is tree sacks (25%), six sacks (50%), nine sacks (75%), and twelve sacks (85%). The isolates characterization has done to the chosen isolates and 14 collected isolates based on their morphology and physiology characteristics. The morphology characteristic are colony shape, structure and Gram stain. The physiology characteristic are qualitative analysis of phosphatase dissolution. Protein reorganization, IAA hormone, deaminase ACC, siderophore, and biological agent ability (producing chitin, HCN and ammonia). The quantitative analysis has done to examine the concentration of produced IAA hormone. The characteristic resulted in the 11 most promising candidate of isolates based on the number of populatin and ability rate was identified microscopically, macroscopically and using moleculer identification. From the 11 isolate, five isolate detected as pathogen bacteria from the Klebsiella genus. The other six isolate were SP 3.7, FH 1.5, K9F.1, K9F.3, K9F.6 and K12F.3. The six isolate significantly increase the length, the wet and dry weight of the root and chlorophyll of pak choi crop. Amongst, four antagonist isolate significantly inhibit pathogen growth where the spout growth remains in an optimun condition.
