Abstract

The use of natural ingredients as medicine is common. The stability of natural materials is easily disturbed, especially oxidation that often occur and damage substances contained in these natural materials. Therefore, a polymer-based suspension was made by reducing the particle size to submicroscopic in order to increase the stability of the natural material used. Purple sweet potato leaves contain compounds such as flavonoids, saponins, and tannins, depending on geographic location. This study aims to determine the effect of variations in the concentration of PVA in the formulation and to take flavonoids as active substances. Submicro suspension was made using the Emulsion Solvent Evaporation method. The study used 3 formulas to take the optimum formula based on the %EE and then carried out particle characterization as well as solubility and physical stability testing. The results of phytochemical screening showed that the extract contained steroids, saponins, polyphenols, tannins, and flavonoids. The level of flavonoids in the extract was 177.25 mg / g. The extract has a moisture content of 7.33%; drying shrinkage of 3,80%; and total ash content of 4.33%. Formula 1, 2 and 3 have a %EE of 88.86; 91.89; 89.31%. The pH of each formula is 7.57; 7.65; and 7.63. The characterization results showed that the particle diameter was 165.53 nm; PDI 0.386; and zeta potential of -31.87 mV. The solubility test results showed that the submicroscopic suspension was completely soluble in aquadest and the acidic pH solvent, namely 5% HCL and SGF. Meanwhile, the alkaline solvents, namely 5% NaOH, 5% NaHCO3, and SIF have solubility that is slightly insoluble in the medium. The results of the physical stability test showed that the color changed to a cloudy yellow and faded, then experienced aggregation with the formation of clots and deposits irreversibly, the smell did not change, and the pH decreased from 7.65 to 6.53 on the 12th day. Based on the data obtained, formula 2 is categorized as the optimum formula and the submicro suspension has good particle characteristics but lacks physical stability.
