Abstract

ABSTRACT : Cellulase enzymes are enzymes that play a role in breaking down complex polymers of cellulose into simple monomers of glucose. Cellulase enzymes can be produced by cellulolytic fungi isolated from various types of substrates, one of which is acacia litter which has high cellulose content. Cellulase enzymes are enzymes that are widely developed in various industrial activities. To meet the needs of these enzymes, it is necessary to know fungi that have a high ability to degrade cellulose, so that in the future they can produce cellulase enzymes in larger quantities. This study aims to determine the genus of fungi that have the highest cellulase activity isolated from acacia litter. The methods used in this research are isolation and purification of fungi, qualitative test of cellulase activity, testing of the ability to produce cellulase quantitatively, manufacture of glucose standard curves with DNS method, quantitative test of cellulase activity with DNS method, characterization and identification of fungi that have activity values. highest cellulase. The conclusion of this study was that 75% of fungal isolates isolated from Acacia (Acacia mangium Willd.) plant litter had the ability to produce cellulase enzymes, which amounted to 9 isolates from 12 isolates of fungi. Based on the quantitative test, the fungal isolate that had the highest cellulase activity value was SL5A2 isolate with a cellulase activity value of 13.05 u/ml. and identified as Aspergillus section Circumdati. Keywords: Cellulolytic Fungi, Acacia Litter, Cellulase, Cellulose ABSTRAK : Enzim selulase merupakan enzim yang berperan dalam menguraikan polimer kompleks selulosa menjadi monomer sederhana glukosa. Enzim selulase dapat diproduksi oleh fungi selulolitik yang diisolasi dari berbagai jenis substrat salah satunya serasah akasia yang memiliki kadar selulosa yang tinggi. Enzim selulase termasuk enzim yang banyak dikembangkan dberbagai kegiatan industri. Untuk memenuhi kebutuhan enzim tersebut maka perlu diketahui fungi yang mempunyai kemampuan tinggi dalam mendegradasi selulosa, sehingga kedepannya dapat menghasilkan enzim selulase dalam jumlah yang lebih besar. Penelitian ini bertujuan untuk mengetahui genus fungi yang memiliki aktivitas selulase paling tinggi yang diisolasi dari serasah akasia. Adapun metode yang digunakan pada penelitian ini yaitu isolasi dan pemurnian fungi, uji aktivitas selulase secara kualitatif, pengujian kemampuan menghasilkan selulase secara kuantitatif, pembuatan kurva standar glukosa metode DNS, uji aktivitas selulase secara kuantitatif dengan metode DNS, karakterisasi dan identifikasi fungi yang memiliki nilai aktivitas selulase paling tinggi. Kesimpulan dari penelitian ini diantaranya sebesar 75% isolat fungi yang diisolasi dari serasah tumbuhan Akasia (Acacia mangium Willd.) memiliki kemampuan dalam menghasilkan enzim selulase yaitu berjumlah 9 isolat dari 12 isolat fungi. Berdasarkan uji kuantitatif isolat fungi yang memiliki nilai aktivitas selulase tertinggi yaitu isolat SL5A2 dengan nilai aktivitas selulase sebesar 13,05 u/ml. dan teridentifikasi sebagai Aspergillus section Circumdati.
