Abstract

Artocarpus altilis (daun sukun) termasuk famili Moraceae. Analisa dengan kromatografi lapis tipis menunjukkan reaksi positif terhadap senyawa fenolik dan flavonoid. Penelitian ini bertujuan untuk mengetahui efek imunomodulator ekstrak etanol daun sukun pada tikus putih jantan yang diinduksi Salmonella thypimurium. Parameter imunitas yang diamati yaitu jumlah sel T CD4, leukosit, limfosit, neutrofil, monosit, makroskopik dan mikroskopik limpa. Hewan uji dibagi menjadi 6 kelompok perlakuan yaitu kelompok normal (0,5 mL/kgBB Na CMC), kontrol negatif (0,5 mL/KgBB Na CMC), kontrol positif (Stimuno®), dan kelompok pemberian ekstrak dengan dosis terapi 200 mg/KgBB, 400 mg/KgBB, dan 600 mg/KgBB. Penginjeksian Salmonella thypimurium dilakukan terhadap semua kelompok perlakuan kecuali kelompok normal. Penginjeksian dilakukan secara intraperitoneal pada hari ke-8 dengan dosis 0,5 x 105CFU/mL. Total fenolik dan flavonoid yang terkandung dalam ekstrak etanol daun sukun secara berturut-turut sebesar 294,16 ± 0,442 mgGAE/g ekstrak dan 129,13 ± 0,115 mg/g ekstrak. Hasil pengujian menunjukkan adanya perbedaan yang nyata pada penurunan jumlah limfosit, monosit, neutrofil dan leukosit (p<0,05) serta menunjukkan jumlah sel T CD4 yang sama dengan kelompok normal. Pemberian ekstrak etanol daun sukun pada dosis 400 mg/KgBB dan 600 mg/KgBB dapat menyebabkan hiperplasi pada organ limpa.
