Abstract

Uji Aktivitas Hepatoprotektor Ekstrak Etanol Kulit Buah Cempedak (Artocarpus champeden) Terhadap Kerusakan Hati Pada Tikus Putih Jantan Galur Wistar Terinduksi CCl4 Peggy Yulianda Jurusan Farmasi FMIPA Universitas Sriwijaya Abstract Cempedak peel is one of the plants that has the potential to be a source of antioxidants as an antidote to free radicals. The skin of cempedak peel ethanol extract contain secondary metabolites flavonoids, saponins,tannins and steroids where the secondary metabolit compouds are thought to be efficacious as hepatoprotectors. This study aims to determine the activity of ethanol extract of cempedak peel as a hepatoprotector through measurement of SGPT and SGOT levels as well as histopathological picture and macroscopic picture induced with CCl4. The mice were divided into six groups, namely the normal group (Na CMC 1%), the positive group (Silimarin 200 mg/kgBB), the negative group (CCl4 0.1 mL/kgBB) as well as the treatment group of ethanol extract of cempedak peel at a dose of 250,500, 1000 mg/kgBB. Variations in the dose of ethanol extract of the skin of cempedak fruit had an effect in decreasing levels of SGPT and SGOT in male white rats wistar-induced strains CCl4. The larger the feeding dose the greater the effect of decreasing SGPT levels and SGOT levels. Based on the results of positive group post hoc analysis with test III there were no significant differences. The results of the picture of histopaatology hepar normal group, positive, negative, test I, test II and test III in the form of necrosis conditions in a row 0, 1, 3, 2, 1, 1. Macroscopic image measurement of liver organs in test group III in accordance with normal liver criteria. Based on the percentage relationship of hepatoprotector effect to the dose, the value of ED50 cempedak peel ethanol 232,76 mg/kgBB. Keywords : Artocarpus champeden, hepatoprotector, SGPT, SGOT, histopathology, macroscopic liver Abstrak Kulit buah cempedak merupakan Salah satu tanaman yang berpotensi untuk menjadi sumber antioksidan sebagai penangkal radikal bebas.kulit buah cempedak dalam ekstrak etanol mengandung senyawa metabolit sekunder seperti flavonoid saponin, tannin, steroid dimana senyawa tersebut diduga dapat berkhasiat sebagai hepatoprotektor. Penelitian ini bertujuan untuk mengetahui aktivitas ekstrak etanol kulit buah cempedak sebagai hepatoprotektor melalui pengukuran kadar SGPT dan SGOT serta gambaran histopatologi dan gambaran makroskopis yang diinduksi dengan CCl4. Tikus dibagi menjadi enam kelompok, yaitu kelompok normal (Na CMC 1%), kelompok positif (Silimarin 200 mg/kgBB), kelompok negatif (CCl4 0,1 mL/kgBB) serta kelompok perlakuan ekstrak etanol kulit buah cempedak dengan dosis 2500, 500, 1000 mg/kgBB. Variasi dosis ekstrak etanol kulit buah cempedak berpengaruh dalam penurunan kadar SGPT dan SGOT pada tikus putih jantan galur wistar yang diinduksi CCl4. Semakin besar dosis makan semakin besar efek penurunan kadar SGPT dan kadar SGOT. Berdasarkan hasil analisis post hoc kelompok positif dengan uji III tidak terdapat perbedaan yang signifikan. Hasil gambaran dari histopaatologi hepar kelompok normal, positif, negatif, uji I,uji II dan uji III berupa kondisi nekrosis secara berturut 0, 1, 3, 2, 1, 1. Pengukuran gambar makroskopis organ hati kelompok uji III sesuai dengan kriteria hati normal. Berdasarkan hubungan persentase efek hepatoprotektor terhadap dosis, maka nilai ED50 ekstrak etanol kulit buah cempedak yaitu 232,76 mg/kgBB.
