Abstract

Antibacterial Assay of Submicro Particles Chitooligosaccharide Against Staphylococcus aureus and Escherichia coli In Vitro Ropiana Purwaningsih 08061181722013 ABSTRACT Chitooligosaccharide is a derivative of chitosan resulting from the deacetylation of chitin and it has been widely reported that Chitooligosaccharide has been shown to have biological activity as an antibacterial. The purpose of this research is to make a submicroparticle formulation of Chitooligosaccharide using sodium alginate polymer. Variations in concentration of sodium alginate were carried out to determine the characteristics of the resulting Chitooligosaccharide submicroparticles. This research begins with the preparation of materials followed by the manufacture of Chitooligosaccharide submicroparticles. Ionic gelation method was chosen in the manufacture of submicroparticles with components consisting of Chitooligosaccharide, sodium alginate and CaCl2 as a crosslinker using variations of sodium alginate 1.6; 3.2 and 4.8 mg. The best formula submicroparticles were characterized by PSA to determine the particle size. The best formula for submicroparticles has a percent encapsulation efficiency of 90.72% with a particle size characteristic of 556.8 nm; PDI value is 0.540 and zeta potential is +36.1 mV. The FTIR spectra of submicroscopic Chitooligosaccharide particles did not show new peak formation or peak reduction, meaning that there was no chemical interaction between Chitooligosaccharide and sodium alginate. So that chitooligosaccharide can be formulated in the preparation of submicroparticles with sodium alginate polymer. Furthermore, antibacterial testing was carried out by the agar diffusion method with disc paper. The results showed that 0.5% Chitooligosaccharide Submicroparticles had antibacterial activity against Staphylococcus aureus and Escherichia coli with inhibition zones of 21 ± 1 mm (very strong zone) and 26 ± 2 mm (very strong zone).
