Abstract
One of the heavy metal pollution in rivers comes from industrial waste and will accumulate in aquatic biota. Efforts to overcome it through the process of phytoremediation using mangrove plant S. caseolaris. The phytoremediation process of heavy metals can cause stress for S. caseolaris and increase free radicals. Adaptation response S. caseolaris in that involve changes in antioxidant enzyme activity including peroxidase (PO), polyphenol oxidase (PPO), and catalase (CAT) so a study was conducted to determine the activity of antioxidant enzyme PO, PPO, dan CAT as adaptation response S. caseolaris in phytoremediation of heavy metals in Pulau Payung, South Sumatra. The sampling process uses the method convenience sampling. Measurement of heavy metal levels of Pb and Cu using atomic absorption spectrophotometry method. Protein extraction using the principle refreegerated centrifuge. Measurement of total protein content with Biuret reagent Biosystem 2000. Determination of activity of PO with hydrogen peroxide and pyrogalol as substrates; PPO with pyrogalol substrate; and CAT with hydrogen peroxide as substrate using uv-vis spectrophotometry method with a wavelength for each enzyme 420 nm; 420 nm; and 240 nm. Analysis of the data used in the form of quantitative data for measuring parameters of pH and temperature, levels of Pb and Cu, as well as activity data for PO, PPO, and CAT presented by analysis of the average data center and standard deviation. Based on the research that has been done, the antioxidant enzyme activities include PO, PPO, and CAT in S. caseolaris in phytoremediation of heavy metals Pb and Cu in Pulau Payung each obtained with PO activity 45,78 U/mg protein/minute; PPO activity 109,05 U/mg protein/minute; and CAT activity 32,02 U/mg protein/minute. The activity of antioxidant enzymes PO, PPO, and CAT is a adaptation response S. caseolaris caused by accumulate (phytoremediation) of heavy metals Pb and Cu in sediment and serves to prevent Reactive Oxygen Species (ROS) formed. Pencemaran logam berat di sungai salah satunya berasal dari limbah industri dan akan terakumulasi pada biota perairan. Upaya untuk mengatasinya melalui proses fitoremediasi menggunakan tumbuhan mangrove S. caseolaris. Proses fitoremediasi logam berat dapat menyebabkan cekaman bagi S. caseolaris dan meningkatkan radikal bebas. Respons adaptasi S. caseolaris berupa perubahan aktivitas enzim antioksidan meliputi peroksidase (PO), polifenol oksidase (PPO), dan katalase (CAT) sehingga dilakukan penelitian yang bertujuan untuk mengetahui aktivitas enzim antioksidan PO, PPO, dan CAT sebagai respons adaptasi S. caseolaris dalam fitoremediasi logam berat di Pulau Payung Sumatera Selatan. Proses pengambilan sampel menggunakan metode convenience sampling. Pengukuran kadar logam berat Pb dan Cu menggunakan metode spektrofotometri serapan atom. Ekstraksi protein menggunakan prinsip refreegerated sentrifuge. Pengukuran kadar protein total dengan reagen Biuret Biosystem 2000. Penentuan aktivitas dari PO dengan substrat hidrogen peroksida dan pirogalol; PPO dengan substrat pirogalol; dan CAT dengan substrat hidrogen peroksida menggunakan metode spektrofotometri uv-vis dengan panjang gelombang pada masing-masing enzim berturut-turut 420 nm; 420 nm; dan 240 nm. Analisis data yang digunakan berupa data kuantitatif untuk pengukuran parameter pH dan suhu, kadar Pb dan Cu, serta data aktivitas PO, PPO, dan CAT yang disajikan dengan analisis pemusatan data rata-rata dan standar deviasi. Berdasarkan penelitian yang telah dilakukan, aktivitas enzim antioksidan meliputi PO, PPO, dan CAT pada S. caseolaris dalam fitoremediasi logam berat Pb dan Cu di Pulau Payung diperoleh masing-masing dengan aktivitas PO 45,78 U/mg protein/menit; aktivitas PPO 109,05 U/mg protein/menit; dan aktivitas CAT 32,02 U/mg protein/menit. Aktivitas enzim antioksidan PO, PPO, dan CAT merupakan respons adaptasi S. caseolaris yang disebabkan oleh pengaruh akumulasi (fitoremediasi) logam berat Pb dan Cu pada sedimen dan berfungsi menangkal Reactive Oxygen Species (ROS) yang terbentuk.
