Abstract
Background: SARS-CoV-2 is the leading cause of world health problems today. Effective vaccines can prevent and treat a disease. The effectiveness of the vaccine can be seen from the antibody titers formed after vaccination. TNF-α is a powerful cytokine that plays a role when inflamed by antigen exposure and indirectly helps B cells to make antibodies. Polymorphism in TNF-α promoters affects the amount of TNF-α secretion, which indirectly also affects the effectiveness of a vaccine. The purpose of this research was to find out the distribution of polymorphism at the point of -308 TNF-α and the characteristic antibody titers of Sriwijaya University employees after the vaccination of the second dose of SARS-CoV-2 inactivated vaccine. Method: This research was an observational descriptive study. The sample was taken from employees of Sriwijaya University who had carried out the second SARS-CoV-2 vaccination in the period March-August 2021. Furthermore, antibody titers examination with CMIA method and genotype examination with PCR-RFLP method. Result: There were 25 identifiable samples. The results of the genotype research found GG genotypes as many as 18 samples (72%), AG 4 samples (16%), and AA 3 samples (12%). In the categorization of IgG antibody titers were obtained a low category of 15 samples (60%) and a high category of 10 samples (40%). In the GG genotype there were 7 samples with low category IgG titers, AG genotypes balanced between high and low categories, and genotype AA 2 people with high category IgG titers. Conclusion: The majority of the samples had homozygous wildtype genotype (GG) and most of the samples had low-category IgG antibody titers after the second dose of SARS-CoV-2 inactivated vaccine vaccination.
