Abstract

Flavonoids is one of the secondary metabolites of breadfruit leaves (Artocarpus altilis (Park.) Fosberg) which plays a critical role in wound healing. The breadfruit leaf extract was made in the form of transdermal drug delivery system purposes for delivery of the drugs through the skin into the systemic circulation in the form of a transdermal film. The polymers form an important component in transdermal film formation, thereby controlling the release of breadfruit leaf extract. The aim of this research were to study the effect of variations in the concentration of chitosan polymer to characteristics of the transdermal film which preparations were formulated with variations in the concentration of chitosan polymer by 1%; 2%; and 3%. Transdermal film’s evaluation included organoleptic, folding endurance, thickness of the film, mechanical properties, water vapor transmission rate, swelling degree, release profil of drugs from the film, spectroscopy FTIR, and morphological forms. The results showed that the film had the green color, distinctive smell, and flexible. The increased concentration of chitosan polymer had a significant effect (p < 0,05) on film thickness,elongation of break , water vapor transmission rate, swelling degree and has no significant effect (p > 0,05) on the tensile strength. The best penetration of drug was showed by formula 1 film with % penetration was 32,61% and flux was 37,55 μg cm-2 / hour-1. FTIR analysis of the best formula showed the occurrence of cross-linked chitosan-tripolyphosphate is marked by the formation of new peak at a wavenumber of 1556,26 cm-1 and transdermal film had a good morphological homogeneity of the film surface dan fine textured
