Abstract
Research on the activity of extracellular azoreductase enzymes from Bacillus tropicuss BD 01 has been carried out. Extract of azoreductase enzyme from bacteria B. tropicuss BD 01 was produced by centrifugation process of bacterial liquid culture at 3000 rpm and 4°C. The crude extract of the enzyme was then fractionated with ammonium sulfate and dialyzed. The results of the enzyme fraction with the highest specific activity were used to determine the optimum conditions for the extracellular azoreductase enzyme from B. tropicus BD 01 bacteria with variations in incubation time, pH, incubation temperature and substrate concentration. Enzyme activity assay was determined using the UV-Vis spectrophotometry method. The enzyme fraction with the highest specific activity was produced in fraction 3 at 40-60% ammonium sulfate saturation with a specific enzyme activity value of 0.00013 mg/mL, an enzyme activity of 0.0254 U/mL, and a protein content of 18.4 U/mg. Optimum conditions The highest azoreductase enzyme activity was at an incubation time of 5 minutes, pH 7, temperature 37°C and substrate concentration of 70 M with an enzyme activity value of 0.1303 U/mL. The extracellular azoreductase enzyme kinetics test from B. tropicus BD 01 bacteria against methyl red dye resulted in a Vmax value of 0.1526 U/mg and a Km value of 11.0745 M.
