Abstract

Penelitian ini dilaksanakan dari bulan Agustus sampai dengan Oktober 2010, bertempat di Laboratorium Fisiologi Hewan, Jurusan Biologi, Fakultas Matematika dan Ilmu Pengetahuan Alam, Universitas Sriwijaya, Inderalaya. Tujuan dari penelitian ini untuk mengetahui efek fraksi etanol daun gandarusa terhadap penurunan kuantitas dan kualitas spermatozoa mencit. Mencit jantan berjumlah 25 ekor dengan berat 30-40 g berumur 3 bulan dikelompokkan menggunakan rancangan acak lengkap (RAL) dengan 5 ulangan untuk masing-masing perlakuan. Perlakuan yang digunakan pada penelitian ini, yaitu P0 (Akuades), P1 (2,23 mg/10 g BB), P2 (4,46 mg/10 g BB), P3 (8,92 mg/10 g BB), dan P4 (17,94 mg/10 g BB) yang diberikan secara oral gavage selama 35 hari. Pada hari ke-36 mencit didislokasi lehernya kemudian sepasang epididimis diambil untuk dijadikan suspensi sperma dalam larutan NaCl 0,9 %, selanjutnya dilakukan pengamatan kuantitas (jumlah) spermatozoa dan kualitas spermatozoa yang meliputi motilitas, viabilitas dan morfologi spermatozoa mencit. Data dianalisis dengan Analisis of Varians (ANOVA) dan dilanjutkan dengan uji Duncan’s Multiple Range Test (DMRT). Hasil penelitian menunjukkan bahwa penurunan kuantitas sperma mencit sangat pada(p <0,01) dosis 17,84 mg/10 g BB, penurunan motilitas sperma mencit sangat signifikan dimulai darisignifikan dosis 4,46 mg/10 g BB, serta penurunan viabilitas dan morfologi normal sperma mencit sangat signifikan dimulai dari dosis terendah 2,23 mg/10 g BB.
