Abstract

The Sumatran elephant (Elephas maximus sumatranus Temminck, 1847) is one of the endemic animals protected. The Elephant Training Padang Sugihan is a part of the conservation in South Sumatran, there are 31 Sumatran elephants consisting and total the birth rate of elephants with 11 calves. The genealogical data of elephants have not been recorded optimally. Thus, phylogenetic analysis is needed. Phylogenetic analysis based on the mitochondrial D-loop region aims to determine the relationship. This research used a non-invasive method in the form of fecal samples. The MDL3 and MDL5 primers used successfully amplified region D-loop mtDNA in 27 samples out of a total of 31 samples. Total range of base lengths in the 27 samples that were successfully amplified was as expected, namely 633 bp to 705 bp. The alignment results showed that elephants in this Elephant Training Center had low nucleotide variations. The similarity of nucleotide sequences was reflected in the genetic distance with genetic distance values in the ingroup group or sample of 0.000% and 0.003%. The lower the value of the genetic distance, the closer the kinship between individuals. The ingroup was more closely related to the outgroup EF588275.2 Asian elephants from Thailand and KX027533.1 Mammoth, with a mean genetic distance value of 0.4% compared to the AF219240.1 African elephant. Reconstruction of phylogenetic trees revealed that Sumatran elephants at the Elephant Training Center of Padang Sugihan were closely related between individuals grouped in the same sub-cluster with an average similarity between individuals of 100% based on genetic distance values and nucleotide sequence similarities in the mitochondrial D-loop region.
