Abstract

Penelitian tentang “Variasi konsentrasi sumber karbon dan nitrogen pada produksi biosurfaktan oleh Pseudomonas alcaligenes sebagai agens bioremediasi minyak bumi” telah dilaksanakan pada bulan Desember 2011 sampai dengan bulan Maret 2012, bertempat di Laboratorium Mikrobiologi, Jurusan Biologi, Fakultas Matematika dan Ilmu Pengetahuan Alam, Universitas Sriwijaya. Penelitian ini bertujuan untuk mengetahui produksi biosurfaktan dengan variasi konsentrasi sumber karbon (crude oil) dan sumber nitrogen (yeast extract dan urea) oleh Pseudomonas alcaligenes sebagai agen bioremediasi minyak bumi. Variasi konsentrasi crude oil yang digunakan yaitu tanpa crude oil, 0,5 g, 1 g, dan 1,5 g. Variasi konsentrasi sumber nitrogen (urea dan yeast extract\ yaitu tanpa nitrogen, 0,1 g, 0,2 g, dan 0,3 g. Penelitian ini menggunakan Rancangan Acak Kelompok (RAK) Faktorial dengan 28 perlakuan dan 2 ulangan. Data dianalis menggunakan Anava (a :5%) dan uji lanjut Beda Nyata Jujur (BNJ). Hasil penelitian menunjukkan bahwa produk biosurfaktan fraksi eksopolisakarida (EPS) yang tinggi didapatkan pada perlakuan CiNj (0,5 g crude oil + 0,1 g yeast extract\ yaitu 2,38 g/L dengan jumlah sel bakteri 1,50.106 cfu/mL dan indeks emulsifikasi 0,60. Produk biosurfaktan fraksi EPS paling rendah didapatkan pada perlakuan C0N0 (tanpa crude oil dan tanpa nitrogen) yaitu 0,36 g/L dengan jumlah sel bakteri 9,01.109 cfu/mL dan indeks emulsifikasi 0,71.
