Abstract

Petai pod is one of the plant’s part that has a potency as antioxidants. Microparticles preparation of pod petai extract aims to deliver pod petai extract targetedly for Alzheimer’s disease. The microparticles is prepared with gelation ionic method using chitosan, sodium tripolyphosphate, and calcium chloride. Formulation of the microparticles preparation of pod petai extract is done by Box-Behnken design in Design Expert®. The factors analyzed to get the optimum formula of the microparticles preparation of petai pod extract are the concentration of chitosan, the concentration of crosslinker, the type of crosslinker, and the speed of the stirring. The optimum formulas obtained are 0.1% of chitosan with 0.233% of STPP and 1% of chitosan with 0.1% of CaCl2. The optimum formula with chitosan-STPP matrix has organoleptic responses such as forming an easily dispersed precipitation on the first day and 45th day; pH 6.103; 84.315% of EE percentage; 71.0846 ppm of petai pod extract concentration after thermodynamic stability testing; 0.2648 ml of sedimentation volume; 1669.667 second of sedimentation time; and one redispersion. The microparticles with chitosan-CaCl2 matrix has organoleptic responses such as not forming any sediment on the first day but precipitating after 45 days; pH 5.87; 92.927% of EE percentage; 87.2836 ppm of petai pod extract after thermodynamic stability testing; 0.08 ml of sedimentation volume; and one redispersion. Physical characterization as in the size of the microparticles of petai pod extract with chitosan-STPP matrix is bigger than the size of microparticles with chitosan-CaCl2 matrix. The particle size’s distribution of the microparticles with chitosan-CaCl2 matrix is bigger than the microparticles with chitosan-STPP matrix. FTIR analysis shows no interaction between petai pod extract with the microparticles matrix as no new group function is detected on the IR spectrum. Intranasal characterization shows that the average weight of each spray is 0.0961 g on the microparticles with chitosan-STPP matrix and 0.1313 g with chitosan-CaCl2 matrix. Based on this result analysis, extract pod petai microparticles could be used in intranasal drug delivery.
