Abstract

Ethanol extract of Vernonia amygdalina Del. leaves is formulated in transfersome dosage form to increase penetration of hydrophilic drugs in the stratum corneum. The transfersome preparation was formulated using lecithin and tween 80 as factor with 32 factorial designs in Design Expert®10 program. The effect of factor compositions and interaction were observed based on the results of the % EE, concentrations, PDI response before and after stability, and the pH from heating cooling cycle stability. Test results were then analyzed with DX®10 to obtain the optimum formula. Optimum formula is obtained with the concentration of lechitin and tween 80 of 92.5 mg dan 10 mg resfectively with desiability 0.769. Factors of lecithin can increase %EE ,concentration, and decrease PDI and pH, while factor tween 80 can decrease %EE, concentration, PDI and pH. The resulting %EE, PDI (poly dispersity index), diameter, and zeta potential analysis of the optimum formula were 83.02 ± 0.03%, 0.4, 342.2 nm, and 121.6 mV. The amount of flavonoid compound in optimum formula is 58.598 ± 0.022 mg and flavonoid in pure extract content is 70,58 mg /0,5 g extract. Stability assay of the grade by heating cooling method with the temperature of 4 and 40oC for three cycles showed that the optimum formula of transfersome more stable than pure extract. Measurement of difused percent of optimum formula and pure extract at 360 minutes was 45.148 ± 0.022% and 24.580 ± 0.022% respectively, which showed greater transfersome penetration ability than African leaf ethanol extract in carrier fluid. The compartement analysis using WinSAAM® program showed that optimum formula transfersomeand african leaf ethanol extract followed 2 compartment model (non lag time) with correlation between Qo and Qc proven by p-value of <0.05. These result prove that either optimum formula transfersome or african leaf ethanol extract is directly distributed to the blood circulation. Based on the research, ethanol extract of leaves Vernonia amygdalina Del. suitable to be made into transfersome because it is more stable and more easily diffused compared with pure extracts.
