Abstract

Diabetes mellitus is a serious chronic disease because the pancreas doesn’t produce enough insulin or the body cannot use insulin effectively. Salam leaves (Syzygium polyanthum Wight.) is a plant containing secondary metabolites of flavonoids, saponins, tannins and phenolics with antidiabetic activity. The aims of the study were to determine the effect of ethanol fraction of salam leaves to decrease blood glucose levels in type 2 diabetes mellitus rats induced by high fat and fructose diet, to determine the ED50 of the ethanolic fraction salam leaves and to see the histopathological description of the pancreas. Induction of high fat and fructose diet was done by giving fructose 1800 mg/kgBW and high fat 15 g/rat (15% cooking oil and 5% duck egg yolk) for 30 days. The ethanol fraction of salam leaves was made with doses variations of 250, 500, and 1000 mg/kgBW which were tested on 6 groups of wistar rats. The positive control group was given metformin at a dose 150 mg/kgBW, the negative control group was given 0.5% Na CMC suspension and the normal group was not given any treatment. The results showed that the % PKGD in the positive control group was 73.83% and the 3 treatment groups with the ethanol fraction of salam leaves at doses of 250, 500, 1000 mg/kgBW, respectively 63.15%, 67.89% and 89.59%. This shows that the ethanol fraction of salam leaves has antidiabetic activity, and there is no significant difference with metformin (p>0.05). ED50 the ethanol fraction of salam leaves is 211.53 mg/kgBW, which is greater than the dose of metformin used, which is 150 mg/kgBW. Pancreatic histopathology results showed improvements in pancreatic β cells by the positive control group and the treatment group at doses of 250, 500 and 1000 mg/kgBW compared to other groups.
