Abstract

The potential utilization of usnic acid is still very limited due to its low solubility in water causes low bioavailability, so that its therapeutic effectiveness also decreases. Chitosan-Alginate polymer based nanocarrier formulation uses ionic gelation method as a new delivery system, so that it would facilitate the absorption process and increase the effectiveness of the usnic acid to achieve the target of treatment. This research aims to obtain the best nanocarrier formula that has good characterization. This research characterizes usnic acid isolate. The study used 3 formulas with various concentration of CaCl2 20 µL, 40 µL, and 100 µL. The optimum formula will be determined based on the highest %EE value and also carried out particle characterization and stability test. The result of the characterization of usnic acid isolate obtained wavelengths of 287 nm in water medium and 230 nm in methanol medium, melting point 204,3-204,8oC, and TLC with Rf 0,42. The first, second, and third formulas had %EE respectively of 95,89%; 96,43%; 98,49%. The characterization results showed the particle size value of 246,3 nm; PDI 0,412; and zeta potential +26,93 mV. Stability test showed that usnic acid nanocarrier in three storage conditions with release model following zero orde, activation energy 6,28 cal/mol, kinetic constant 0,083 mg/days with shelf-life 59 days at temperature 25oC and estimated shelf-life 71 days at temperature 5oC. The results of one-way ANOVA statistical analysis showed a significant effect of CaCl2 variation on the entrapment efficiency and usnic acid nanocarrier increased solubility compared to pure usnic acid (p<0,05). It can be concluded that the usnic acid nanocarrier has good characterization and stability. Keywords : Usnic acid, Chitosan, Natrium Alginate, CaCl2, Nanocarrier. Pemanfaatan potensi asam usnat masih sangat terbatas dikarenakan kelarutannya yang rendah dalam air yang menyebabkan bioavailabilitasnya menjadi rendah, sehingga efektivitas terapinya juga menurun. Formulasi nanocarrier berbasis polimer Kitosan-Alginat menggunakan metode gelasi ionik sebagai sistem penghantaran baru, sehingga akan memudahkan proses absorbsi dan dapat meningkatkan efektivitas asam usnat tersebut untuk mencapai target dari pengobatan. Penelitian ini bertujuan untuk mendapatkan formula terbaik nanocarrier yang memiliki karakterisasi yang baik. Penelitian ini melakukan karakterisasi isolat asam usnat. Penelitian menggunakan 3 formula dengan variasi konsentrasi CaCl2 20 µL, 40 µL, dan 100 µL. Formula terbaik ditentukan berdasarkan nilai %EE tertinggi kemudian dilakukan karakterisasi partikel dan uji stabilitas. Hasil karakterisasi isolat asam usnat didapatkan panjang gelombang 287 nm dalam medium air dan 230 nm dalam medium methanol, titik lebur 204,3-204,8oC, dan KLT dengan Rf 0,42. Formula 1, 2 dan 3 memiliki %EE berturut-turut sebesar 95,89%; 96,43%; 98,49%. Hasil karakterisasi menunjukkan nilai ukuran partikel 246,3 nm; PDI 0,412; dan zeta potensial +26,93 mV. Pengujian stabilitas menunjukkan nanocarrier asam usnat pada tiga kondisi penyimpanan mengikuti model pelepasan orde-0, energi aktivasi 6,28 cal/mol, konstanta kinetik 0,083 mg/hari dengan masa simpan 59 hari pada suhu 25oC dan perkiraan masa simpan 71 hari pada suhu 5oC. Hasil analisis statistik uji ANOVA satu arah menunjukkan pengaruh variasi CaCl2 yang signifikan terhadap efisiensi penjerapan dan nanocarrier asam usnat meningkatkan kelarutan dibandingkan dengan asam usnat murni (p<0,05). Dapat disimpulkan bahwa nanocarrier asam usnat memiliki karakterisasi dan stabilitas yang cukup baik. Kata kunci : Asam usnat, Kitosan, Natrium Alginat, CaCl2, Nanocarrier.
