Abstract
The aim of this study was to see the effect of combination of Lactobacillus acidophilus and Chito-oligosaccharides (COS) on the Growth of Escherichia coli and Staphylococcus aureus. COS 0.2% in 1% acetic acid and 1% acetic acid solution was tested against E. coli and S. aureus. L. acidophilus baeteria before and after combined with COS 0.2% with the variation of time incubation of 44, 46, 48, 50, and 52 hours was tested for antibacterial activity againts E. coli and S. aureus with well diffusion method. Organic acids which was excreted by L. acidophilus after and before combination with COS 0.2% was measured using HPLC method. The results showed that COS 0.2% can inhibit the growth of E. coli and S. aureus with inhibition zone 37.2 mm2 and 52.2 mm2 respectively. Combination of L. acidophilus and COS 0.2% gave inhibition zone greater than L. acidophilus without COS 0.2% againts E. coli and S. aureus. The 48-hours incubation time resulted the greatest inhibition zone againts E. coli and S. aureus were 367.92 mm2 and 343.99 mm2 respectively. Optical density of L. acidophilus combined with COS 0.2% greater than L. acidophilus control. The concentration of organic acids was produced by the combination of L. acidophilus and COS 0.2% lower than the baeteria L. acidophilus without combination with COS 0.2% .
