Abstract

The research about produced cellulase by Aspergillus niger has been done using CMC (Carboxy Methyl Cellulose) substrate. The Identification has been performed according to the growing phase of A. niger, whether the cellulase enzyme is intracellular or extracellular; the optimum conditions of producing cellulase enzyme such as fermentation time, concentration of CMC substrate, and the pH of the medium. The enzyme’s extract is resulted the activity testing with DNS (Dinitrosalisilat) method. Then the enzyme’s extract was ffactinated with ammonium sulphate and done to tested activity and specific activities. From the growing curve, the growing phase of A. niger was obtained which are the 3rd day to the 9th. Based observation of activity intracellular and extracellular showed that enzyme produced by A. niger was extrasellular. The observation of cellulase enzyme activity from A. niger showed that on fermentation time the 10th day with the substrate concentration of 3 % CMC dan medium pH 6, is the optimum condition for producing cellulase enzyme from A. niger with the activity is resulted 0,282 IU/mL. Based on the fractination of cellulase enzyme produced by A. niger resulted 2 ffactions, which are the fractination of 20 - 40 % ammonium sulphate with specific obtained 1,166 IU/mg.
