Abstract

Diuretics are drugs that can increase the rate of urine formation. Cempedak (Artocarpus champeden) contains phenolic compounds potentially as diuretics. This study aims to determine the diuretic activity of the ethyl acetate fraction of cempedak leaves. The total phenolic level of ethyl acetate of the epoxy leaf is determined using a UV spectrophotometer at a maximum wave rate of 744 nm. The diuretic activity test was conducted with 6 testing groups, consisting of normal, negative, positive groups and three dose treatment groups of 25, 50, and 100 mg/200gBW. Observations were made for 24 hours with urine volume parameters and sodium ion levels (Na+), potassium (K+) and chloride (Cl-). The results of the study showed the total phenolic level of the ethyl acetate fraction of the chepid leaves was as large as 71mg GAE/g. Urinary volume of test animals and Na+, K+ and Cl- ions at doses of 100 mg/200gBW showed a real difference with positive control of furosemid characterized by significance values (00.05). The fraction of ethyl acetate at doses of 100 mg/200gBW can potentially be as diuretic as compared with doses of 25 mg/200gBW and 50 mg/200gBW. The higher the dose of the given fraction then the more increased the amount of urine discharge and Na+, K+ and Cl- ion levels. The ED50 value of the ethyl acetate fraction was 23.65 mg/200gBW. Cempedak (Artocarpus champeden) mengandung senyawa fenolik yang berpotensi sebagai diuretik. Penelitian ini bertujuan untuk mengetahui aktivitas diuretik dari fraksi etil asetat daun cempedak. Kadar fenolik total etil asetat daun cempedak ditentukan menggunakan spektrofotometer UV pada gelombang maksimum 744 nm. Uji aktivitas diuretik dilakukan pada 6 kelompok tikus jantan wistar, terdiri dari kelompok normal, negatif, positif dan tiga kelompok perlakuan dosis 25, 50, dan 100 mg/200gBB. Pengamatan dilakukan selama 24 jam dengan parameter volume urin dan kadar ion natrium (Na+), kalium (K+) dan klorida (Cl-). Hasil penelitian menunjukkan kadar fenolik total fraksi etil asetat daun cempedak adalah sebesar 71mg GAE/g. Volume urin hewan uji dan ion Na+, K+ dan Cl- pada dosis 100 mg/200gBB menunjukkan adanya perbedaan nyata dengan kontrol positif furosemid yang ditandai dengan nilai signifikansi (<0.05). Fraksi etil asetat pada dosis 100 mg/200gBB dapat berpotensi sebagai diuretik dibandingkan dengan dosis 25 mg/200gBB dan 50 mg/200gBB. Semakin tinggi dosis fraksi yang diberikan maka semakin meningkat jumlah pengeluaran urin dan kadar ion Na+, K+ dan Cl-. Nilai ED50 dari fraksi etil asetat adalah 23,65 mg/200gBB.
