Abstract
The ethanol extract of mangosteen peels is known to contain xanthones which have anti-inflammatory effects and can accelerate the process of fibroblast proliferation. In addition, xanthones can trigger the formation of collagen which plays an important role in the maintenance of the structure and wound healing. The selection of transdermal patch dosage forms for mangosteen peels extract aims to deliver the active substance through the skin and to the systemic system. An important component in making patches is the use of polymers, so in this study a combination of chitosan and sodium alginate polymer was used. The addition of a crosslinker to the transdermal patch formula can improve the elasticity, stiffness, rigidity of the transdermal patch, and affect the degree of swelling. This study aimed to determine the effect of variations in the concentration of calcium chloride as a crosslinker on the characteristics of the patch. Patches are made by layer by layer method in 3 formulas, namely F1, F2, F3 with a calcium chloride concentration of 0.2; 0.4; 0.6. The best formula was found in the F2’s patch, indicated by a flexible, brownish-colored patch with a smooth surface. In addition, the resulting patch also has a uniform weight, qualified of criteria for a thickness of >300 folds, has a pH of 5 which is safe to use for topical preparations, as well as a low water content of 1.8389%, a swelling degree value of 3.407%, qualified for uniformity of content and the penetration profile of the active substance was 38.87%. The results of the morphological evaluation obtained a pore size of 5,88µm, while the observation of the IR spectrum showed the interaction of cross-linked chitosan, sodium alginate, and CaCl2 to form a new functional group N-C=O at a wavenumber of 1622,23 cm-1. Keywords: Mangosteen peel extract (Garcinia mangostana L.), transdermal patch, and calcium chloride. Ekstrak etanol kulit manggis diketahui mengandung xanthon yang memiliki efek anti inflamasi dan mampu mempercepat proses proliferasi fibroblas. Selain itu, xanthon dapat memicu pembentukan kolagen yang berperan penting dalam pemeliharaan struktur dan penyembuhan luka. Pemilihan bentuk sediaan patch transdermal untuk ekstrak kulit manggis bertujuan untuk menghantarkan zat aktif menembus kulit dan sampai sistem sistemik. Komponen penting dalam pembuatan patch adalah penggunaan polimer, sehingga dalam penelitian ini digunakan kombinasi polimer kitosan dan natrium alginat. Selain itu, penambahan crosslinker pada formula patch transdermal dapat memperbaiki elastisitas, kekakuan, rigiditas dari patch transdermal, dan mempengaruhi derajat swelling. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh variasi konsentrasi kalsium klorida sebagai crosslinker terhadap karakteristik patch. Patch dibuat dengan metode layer by layer dalam 3 formula yakni F1, F2, F3 dengan konsentrasi kalsium klorida 0,2 ; 0,4 ; 0,6. Formula terbaik didapat pada patch F2 ditunjukkan dengan patch yang lentur, berwarna kecoklatan dengan permukaan yang halus. Selain itu, patch yang dihasilkan juga memiliki bobot yang seragam, memenuhi kriteria ketebalan 300 lipatan, memiliki pH 5 yang aman digunakan untuk sediaan topikal, serta kadar air yang rendah yakni 1,8389%, nilai derajat swelling sebesar 3,407%, memenuhi syarat keseragaman kadar dan profil pelepasan zat aktif terpenetrasi sebesar 38,87%. Hasil evaluasi morfologi diperoleh ukuran por-pori permukaan sebesar 5,88µm, sedangkan pengamatan spektrum IR menunjukkan terjadinya interaksi dari ikatan silang kitosan, natrium alginat dan CaCl2 membentuk gugus fungsional baru N-C=O pada bilangan gelombang 1622,23 cm-1. Kata kunci: Ekstrak kulit manggis (Garcinia mangostana L.), patch transdermal, Kalsium klorida.
