Abstract

Catechins have low membrane permeability and poor absorption in the gastrointestinal track (GIT). SNE can increase the % dissolution of the active substance because the active substance is dissolved in the nano oil globules which are wrapped by a layer of surfactant and cosurfactant. The concentration of SNE components is determined by performing an optimization process assisted by I-optimal mixture design (IOMD) combined with PCA-CA chemometric analysis. The combination of SNE optimization with a chemometric approach aims to evaluate the design and clarify the relationship between the resulting responses. The components used are oleic acid (oil), Tween 80 (surfactant), and propylene glycol (cosurfactant). Evaluation parameters include emulsification time, % transmittance, viscosity, particle size, zeta potential, mobility and polydispersity index. Component interactions were evaluated using the Fourier transform Infrared Spectrophotometry Attenuated Total Reflectance (FTIR-ATR). The characteristics of the optimal SNE formula produced are the SNE system which has a transmittance value of 98.1% and a viscosity value of 7.169 pouses, and nanoemulsions which have an emulsification time of 6.47 seconds (water), 7.46 seconds (SGF), 7.01 seconds. (SIF), % transmittance of water nanoemulsion 93.93%, % transmittance of SIF nanoemulsion 92.37%, zeta potential value -27.63 m/V, mobility -2.165 Nm cm/Vs , particle size 105.2 d.nm, and PDI 0.25. The optimal formulas of oleic acid with a concentration of 10%, Tween 80 with a concentration of 60%, and propylene glycol 30% met the characteristics and had correlation. Keywords: SNE, Catechins, I-optimal mixture design, nanoemulsion. Katekin memiliki permeabilitas membran rendah dan penyerapan yang buruk pada gastrointestinal track (GIT). SNE dapat meningkatan % disolusi zat aktif karena zat aktif terlarut dalam globul minyak nano yang terbungkus oleh lapisan surfaktan dan kosurfaktan. Konsentrasi komponen SNE ditentukan dengan melakukan proses optimasi dibantu dengan I-optimal mixture design (IOMD) dikombinasikan dengan analisa kemometrik PCA-CA. Kombinasi optimasi SNE dengan pendekatan kemometrik bertujuan untuk mengevaluasi desain dan memperjelas hubungan antar respon yang dihasilkan. Komponen yang digunakan berupa asam oleat (minyak), Tween 80 (surfaktan), dan propilenglikol (Kosurfaktan). Parameter evaluasi meliputi waktu emulsifikasi, %Transmitan,viskositas,ukuran partikel,zeta potensial, mobilitas dan indeks polidispersitas. Interaksi komponen dievaluasi menggunakan fourier transform Infrared Spectrophotometry Attenuated Total Reflectance (FTIR-ATR). Karakteristik formula optimal SNE yang dihasilkan yaitu sistem SNE yang memiliki nilai transmitan 98,1% dan nilai viskositas 7,169 pouse, serta nanoemulsi yang memiliki waktu emulsifikasi sebesar 6,47 detik (media air), 7,46 detik (media SGF), 7,01 detik (media SIF), % transmitan nanoemulsi air 93,93 %, % transmitan nanoemulsi SIF 92,37%, nilai zeta potensial -27,63 m/V, mobilitas -2,165 Nm cm/Vs , ukuran partikel 105,2 d.nm, dan nilai PDI 0,25. Formula optimal asam oleat dengan konsentrasi 10%, Tween 80 dengan konsentrasi sebesar 60%, dan propilenglikol 30% memenuhi karakteristik dan memiliki korelasi. Kata kunci :SNE, Katekin, I-optimal mixture design, nanoemulsi.
