Abstract

Excessive free radical activity can damage the health of the body. Free radicals can enter the body and attack healthy cells and cause these cells to lose their function and structure. Cell damage caused by free radicals can cause various degenerative diseases. To prevent the negative effects of free radicals on the body, antioxidant compounds are needed. One of the natural sources that can be used as a source of antioxidants is guava bark (Syzygium aqueum). This study aims to determine the antioxidant activity and compounds isolated from the methanol extract of guava stem bark. The study was initiated by maceration extraction using methanol as a solvent and separation and purification were carried out using chromatographic techniques. The structure of the compound was determined based on the analysis of FTIR and NMR spectroscopic data (1H-NMR and 13C-NMR). Determination of antioxidant activity was carried out by the free radical scavenging method DPPH which was expressed by the IC50 value. The results of the antioxidant activity research showed that the methanol extract of guava stem bark had a very strong antioxidant activity with an IC50 value of 4,10 µg/mL. The isolation results from the methanol extract obtained pure compounds, based on analysis of spectroscopic data and compared with data from the literature showed that the isolated compounds were in the steroid group, namely β-sitosterol. The pure compound obtained from the isolation was tested for antioxidant activity which showed that the isolated compound (isolate A) had a very strong antioxidant activity with an IC50 value of 11,01 µg/mL and the isolated compound (isolate B) did not have antioxidant activity with an IC50 value of 745,37 µg/mL. The smaller the IC50 value, the higher the antioxidant activity of a compound.
