Abstract

Actinomycetes are one of the microorganisms that have commercial value because of their ability to produce antibiotics. 80 % of antibiotics on the market today are produced by Streptomyces. Exploration of Actinomycetes from swampland revealed 19 isolates and 4 isolates of which ACT 8, ACT 10, ACT 11 and ACT 13 had high antibacterial activity against Escherichia coli ATCC25922 and Staphylococcus aureus ATCC25923, based on morphological and biochemical characterization of the four isolates suspected to be genus Streptomyces. In determining the identity of Actinomycetes, microbiologists combined various components such as morphological, biochemical and genomic characters in polyphasic studies. The character of the surface ornamentation of Actinomycetes spores is the key to identify that distinguishes among the species. Identification of Actinomycetes using DNA-barcoding and adding the ornamental character of the spore surface were able to determine the type of Actinomycetes accurately. In this study, antibacterial activity was tested, morphological structure visualization using Scanning Electron Microscope (SEM) and molecular analysis using DNA-barcoding 16S rRNA gene, so the identity of the four isolates of Streptomyces spp. can be known with accurately. Four isolates of Streptomyces spp. had a strong ability to inhibit the growth of gram-negative bacteria (E. coli) while against gram-positive bacteria (S. aureus) ACT 8 had strong abilities, isolates ACT 10, ACT 11 and ACT 13 had moderate abilities. Four isolates had the same hyphal structure with many branches forming a complex mycelium such as root fibers and found sporophores and had many spores with similar shapes, which is called spiral chains. The shape of the spore chain is a characteristic that is found in the genus Streptomyces. Based on the results of homology search, ACT 8 had the highest percent identity against S. samsunensis strain M1463 with a value of 99.72% while isolates ACT 10, ACT 11 and ACT 13 had the highest percent identity against S. malaysiensis strain NBRC 16446 with a value of 99.45 % to 99.79%. Genetic distance analysis and phylogenetic tree reconstruction showed that the four isolates were closely related to Streptomyces samsunensis sp. nov. and Streptomyces malaysiensis sp. nov. with a genetic distance ranging from 0.001 to 0.006 and belongs to the clade Streptomyces violaceusniger
