Abstract

Ikan sumatra (Puntius tetrazona) merupakan salah satu jenis ikan hias air tawar endemik yang tersebar di Pulau Sumatera dan Kalimantan. Ikan ini memiliki beberapa varietas diantaranya tiger barb, green tiger barb, albino tiger barb dan balloon tiger barb. Tujuan penelitian ini untuk mengetahui sekuen gen COI DNA mitokondria dan konstruksi filogenetik. Penelitian ini dilaksanakan pada bulan Oktober-Desember 2021. Metode yang digunakan dalam barcoding DNA spesies ikan sumatra yaitu ekstraksi DNA, amplifikasi DNA menggunakan Polymerase Chain Reaction (PCR), elektroforesis dan sekuensing daerah gen COI pada mtDNA. Sampel ikan didapatkan dari Sungai Lematang, Sungai Musi dan dari budidaya komersil di Palembang. Fragmen gen COI mtDNA yang telah disekuensing didapatkan hasil PCR dengan optimasi suhu annealing 52℃ selama 45 detik dalam 32 siklus (sampel dari Sungai Lematang) dan 51℃ selama 30 detik dalam 35 siklus (sampel dari Sungai Musi dan dari budidaya komersil). Sekuensing gen COI menghasilkan panjang nukleotida 645 base pairs (bp). Hasil analisis BLASTn sampel ikan sumatra asal Sungai Lematang dan Sungai Musi memiliki kemiripan tertinggi sebesar 93,00%-93,30% dengan spesies P. tetrazona sementara sampel asal budidaya komersil Palembang memiliki kemiripan tertinggi sebesar 97,34%-97,60% dengan spesies yang sama pada data Genbank. Filogenetik dua populasi ikan sumatra asal Sungai Lematang dan Sungai Musi terhadap budidaya komersil Palembang membentuk dua subcluster yang terpisah.
