Abstract

The UAE and MAE extraction methods are modified extraction methods for extracting natural compounds.The extraction of cempedak seeds aims to determine the optimal conditions for each method based on the total flavonoid content and IC50 antioxidant activity obtained and perform a meceration extraction based on the results of the optimum conditions. The UAE method research uses variations in solvent concentration (30%, 70%, 96%) and extraction time (20,50,80 minutes) while the MAE method uses variations in power (90,180,360 watts) and variations in extraction time (10,25, 40 minutes). The best conditions were determined using Design Expert® 10. The best UAE extraction conditions were at a concentration of 70% and an extraction time of 80 minutes, while the best MAE conditions were at 180 watts and an extraction time of 25 minutes. It is based on the highest desirability value. UAE extraction using 70% ethanol and extraction time of 80 minutes resulted in flavonoid content of 250,676 mg/100g and antioxidant IC50 value of 35,133 µg/mL. MAE extraction using 180 watts of power and extraction time of 25 minutes resulted in flavonoid content of 230.828 mg/100g and antioxidant IC50 value of 41.255 µg/mL. Meceration extraction with 70% ethanol and extraction time of 25 minutes resulted in flavonoid content of 270.946 ± 0,690 mg/100g and antioxidant IC50 value of 27.548 ± 6,840 µg/mL.
