Abstract

Cempedak fruit skin (Artocarpus integer (Thunb.) Merr.) contains flavonoid compounds that function as antioxidants. Bioactive compounds in plants are obtained by extraction method. UAE and MAE extraction methods were chosen because it use a shorter time and less solvents than conventional methods. The purpose of this research is to determine the optimal conditions for solvent concentration and extraction time on the UAE method and the power and extraction time on the MAE method. This research was conducted by varying the solvent concentration (30, 70, 96%) and extraction time (20, 50, 80 minutes) on the UAE method, and power (90, 180, 360 watt) and extraction time (10, 25, 40 minutes) on the MAE method. Determination of the best extraction conditions was carried out using a factorial design. Based on Design Expert 12® analysis, in the UAE method the factor of solvent concentration has no effect and the extraction time affects the total flavonoid content, but the factor of solvent concentration and extraction time affect the antioxidant activity, and in the MAE method factor of power and time extraction did not affect total flavonoid content and antioxidant activity. The best UAE method conditions use ethanol concentration 70% and extraction time 80 minutes with a total flavonoid content value of 304,558 mg/100g and antioxidant IC50 antioxidant 37,962 µg/mL, and the best condition of MAE at 180 watt and extraction time of 25 minutes with a value of total flavonoid content of 267,568 mg/100g and antioxidant IC50 89,611 µg/mL. The result of maceration with condition ethanol concentration 70% and extraction time 25 minutes obtained total flavonoid 304,307±0,004a mg/100g and antioxidant IC50 28,700±0,263a µg/mL.
