Abstract
Red spinach leaves contain anthocyanins which have pharmacological effects as antioxidants. The purpose of this study was to extract anthocyanin compounds from red spinach leaves using the UAE (Ultrasonic Assisted Extraction) technique. The optimization of ultrasonic extraction of red spinach leaves aims to determine and find out the extraction temperature, extraction time and pH of the best solution in determining the percent yield, total anthocyanin levels, and IC50 antioxidant activity in red spinach leaves. The research was conducted by varying the extraction temperature (25 ; 42.5 ; 60ºC), extraction time (10, 20, 30 minutes), and pH (1, 2, 3). Determination of the best extraction conditions was analyzed using CCD (Central composite design). Time of extraction, temperature of extraction, and pH factors affected the percent of yield, total anthocyanin levels, and IC50 antioxidant activity of red spinach leaves. The best UAE extraction conditions were obtained at an extraction temperature of 45ºC, extraction time of 13 minutes and pH 1 produced 20.694% yield, total anthocyanin content of 348.489 mg/100g, and IC50 of 24,448 g/mL. Based on the characterization test of the extracts under optimum conditions which included moisture content, drying loss, total ash content, the results were obtained that cormformed on requirements of the Indonesian Herbal Pharmacopoeia.
