Abstract

This study aims to determine the effect of differences in extraction methods on the polysaccharide content and antidiabetic activity of the leaf extract of the apu-apu (Pistia stratiotes) leaf extract. This research method was carried out experimentally in a laboratory using an extraction method consisting of 2 levels (high temperature extraction method and ultrasonic assisted extraction (UAE)) and was repeated 3 times. Then the data obtained were analyzed descriptively and performed a different test (Independent Sample t-Test). Parameters of the tests carried out included extract yield, total sugar content test and crude fiber content test as well as analysis of alpha-glucosidase inhibitory activity. The test results showed a significant difference (p <0.05) to the yield using the high temperature method and the UAE. UAE extract has an extract yield of 9.16%±2.14 higher than the high temperature extract. However, the three tests carried out using high temperatures had a total sugar content of 538.29±17.45 mg glucose eq./g dry sample and crude fiber content of 6.49%±1.18 higher than the UAE method. The extract using the high temperature method also had a higher ability to inhibit alpha-glucosidase with an IC50 value of 394.84±10.92 ppm higher than the UAE extract.
