Abstract

Coal mining process in PT. Bukit Asam, Tanjung Enim, South Sumatra using the open pit mining method which is carried out by blasting and stripping the topsoil. The exposed rock will react with water and oxygen and produce acid mine drainage (AMD) which has an acidic pH and high heavy metal solutability. The AMD treatment can use an active method using chemical material and a passive method using phytoremediation in the wetland. AMD phytoremediation process can cause stress in E. dulcis. The physiological response of E. dulcis in phytoremediation AAT is involve antioxidant enzymes including Peroxidase (PO), Polyphenol Oxidase (PPO), and Catalase (CAT). This study aims to determine the activity of antyoxidant enzymes in roots of E. dulcis on phytoremediation of acid mine drainage in PT. Bukit Asam, Tanjung Enim, South sumatra. The sampling method in this study uses convenience sampling. Measurement of metal content of Fe and Mn using AAS. Protein extraction used the principle of centrifugation at a speed of 12,500g and a temperature of 0°C for 20 minutes. Measurement of total protein content using a mixture of 400µl standard protein (Bovin Serum Albumin). 400µl of distilled water and 200µl of biuret reagent, PO and CAT activities used H2O2 as an substrat, PPO without H2O2. measured absorbance value using uv-vis spectrophotometry. Analysis of the data used consist of quantitative data presented with a analysis table and standart deviations. Based on the research the result of activities anzyme antioxidant E. dulcis on fitoremediation of AMD is PO 178.98 U/mg protein/minute, PPO 388.96 U/mg protein/minute and CAT 184.34 U/mg protein/minute. E. dulcis was able to increase the pH from 5.6 to 7.7 and reduce Fe levels from 157.6 mg/l to 5 mg/l and Mn 34.2 mg/l to 15 mg/l in AMD. Measurement of antioxidant enzyme activity can be used as an evaluation of the review of the success of AMD phytoremediation.
