Abstract

Most of society depends on the use of plastic as packaging material. The material most commonly used as a constituent of plastics comes from PET (Polyethylene terephthalate). Dependence on PET-based plastics causes a high increase in landfills due to non-degradability materials. PET can be degraded by physicals and chemicals method, but those methods raise the problem of secondary environmental pollution and high energy consumption. A more environmentally friendly process is needed to process PET through biodegradation involving microorganisms. IsPETase enzyme from Ideonella sakaiensis has a significant impact on the development of PET biodegradation processing. This paper is written using the literature study method by conducting research studies using research journals, books, reports, and scientific literature related to the issues raised, as well as discussing with accompanying lecturers. The idea is written with an analysis of several plastic waste problems in Indonesia combined with logical solutions based on existing literature reviews. Genetic engineering technology, is applied to optimize the potency of the IsPETase enzyme. The IsPETase enzyme was successfully expressed by the Escherichia coli strain Rosetta Gami-B with the PET22b-SPLamB: IsPETase vector where the activity of E. coli can express PETase heterologically by the translation process consisting of the first 25 amino acids from the protein sequence, with a probability of cleavage location of 0.9316 . This makes IsPETase superior when compared to other homologous cutinase enzymes.
